PRP-HO family:

Our solution will be PRP6-HO7, an injectable human-derived recombinant protein very stable in the circulation and with sustained action to treat autoimmune diseases. The primary indication will be the induction and maintenance of clinical remission of adult patients with moderately to severe IBD who have had an inadequate response with, lost response to, or have become intolerant to conventional therapies.

Thus, our asset is postulated as an immunomodulator and not as an immunosuppressant, a key differential aspect that would justify its development as an innovative drug with a “first-in-class” character and a unique mechanism of action. We have established the immunomodulatory action of PRP-HO7 in human inflammatory dendritic cells and macrophages.  In vivo safety and efficacy have been successfully demonstrated in systemic lupus erythematosus (Luque et al. (2019) Kidney Int.), and in inflammatory bowel disease (IBD) and rheumatoid arthritis (unpublished results) mouse models. Owing to the high cost of biologic drug development, we have also developed a novel, simplified and non-glycosylated recombinant analogue, PRP6-HO7 (EP19382910), showing enhanced efficacy, specificity, and scalable, cost-effective protein production.

La nostra solució serà PRP6-HO7, una proteïna recombinant injectable d’origen humà molt estable en la circulació i amb acció sostinguda per a tractar malalties autoimmunes. La indicació principal serà la inducció i el manteniment de la remissió clínica dels pacients adults  que han tingut una resposta inadequada, han perdut resposta o s’han tornat intolerants als tractaments convencionals.

Així, el nostre actiu es postula com a immunomodulador i no com immunosupressor, un aspecte diferencial clau que justificaria el seu desenvolupament com un fàrmac innovador amb caràcter de “primer en la seva classe” i amb un mecanisme d’acció únic. Hem establert l’acció inmunomoduladora del PRP-HO7 en cèl·lules dendrítics inflamatòries humanes i macròfags. La seguretat i eficàcia in vivo han estat demostrades amb èxit en models de ratolí de lupus eritematós sistèmic (Luque et al. (2019) Kidney Int.), i en models de malaltia inflamatòria intestinal (EII) i artritis reumatoide (resultats no publicats). A causa de l’alt cost del desenvolupament de fàrmacs biològics, també hem desenvolupat un nou anàleg recombinant, simplificat i no glicosilado, PRP6-HO7 (EP19382910), que mostra una major eficàcia, especificitat i producció de proteïnes escalable i rendible.
